S95 : Analysis of diversity and genetic relationship of different grape cultivars in Shahrood region baxsed on Retrotransposon-baxsed molecular marker REMAP

Abstarct: Recognition the genetic diversity of grape was helped the breeding of this plant. The purpose of the study is investigation of genetic diversity in 25 different grape variety by using morphological data, retrotransposon markers REMAP and IRAP and also ISSR markers. In order to doing this research, sampling and evaluation of morphological traits was done in Shahrood’s researches center and Sisakht gardens during the summer of 2012. In this study 14 morphological traits related with leaf shape, bunch and berry shape was investigated. In molecular part, 21 retrotransposon and ISSR primers were used. On the basis of bond formation rate, polymorphism and repeatability of bands, 15 primers were selected. This 15 primers produced 156 bands so 144 bands showed acceptable polymorphism. The number of replicated fragments by different primers were varied, the highest number of fragments was 16 belong to "825" and "840" primers and the lowest of them was 6 belong to "vine1fa+Ms3" (REMAP) primer combination and "857" (ISSR) primer. Mean of polymorphic bonds number for each primer, was 9.6 bonds. The highest Shannon's variation coefficient and observed heterozygosis belong to Gret1Fa primer from IRAP primers so that probability indicated IRAP marker with regard to other studied markers was better. Cluster analyses were done using UPGMA algorithm and Jacard similarity matrices. The results of this research indicate a molecular and morphological diversity among grape varieties and also showed the efficiency of retrotransposon markers, REMAP and IRAP, and so ISSR marker for determine genetically diversity of grape varieties that were studied.
