S708 : The genetic diversity of commercial grape varieties by ISSR and RAPD markers

Abstarct: Understanding the genetic diversity of the grape plant helps to eugenic researches of this plant. In this research, the genetic diversity of 25 Iranian grapevine cultivars is evaluated using morphological data and molecular markers of the ISSR and RAPD. This research was carried out on grapevines of research collections belonging to Agricultural Research Center of Bojnourd. In the study of morphological diversity, 21 traits related to vegetative characteristics were evaluated and in the molecular section, 10 ISSR and 10 RAPD primers were used. The results from the 21 traits indicated that there is a large variation in the traits being evaluated. The highest diversity index (99.23%) related to the straight indumentum density trait of the main petals in the lower surface of lamina and the lowest diversity index (12.5%) was related to the length to width ratio of the blade. The results of correlation coefficients show a positive and significant correlation between some of the traits. In the dendrogram, the morphological traits were divided into four distinct groups of 15 cultivars distance. 25 grapevine cultivars with 10 ISSR primers produced a total of 103 bands, of which 98 showed polymorphism. The 10 RAPD primers produced a total of 113 bands in 25 different grapevine cultivars, all of which were polymorphic. The UBC888 marker with 13 bands showed the highest number of bands and the UBC850 marker with 7 bands showed the least number of bands in the ISSR primer. The OPK marker with 14 bands showed the highest number of bands and the TIBM17 marker with 8 bands showed the lowest number of bands in the RAPD primer. The highest resolving power was related to the OPO08 primer (8.64) and the lowest resolving power associated with UBC887 primer (2.56). In cluster analysis with ISSR markers, cultivars were categorized into four groups and grouped by RAPD markers into four distinct groups. The results of both markers showed that two cultivars “Khoshnav” and “yellow Khalili” are at a far longer genetic distance than other cultivars. The results of this study indicate the presence of molecular and morphological diversity among grapevine cultivars.
