S653 : Genom-wide analysis of sulfate transporter (SULTR) gene family in durum wheat and their exxpression study in response to salt stress

Abstarct: Abstract. Sulfate transporters (SULTR) play an important role in the uptake and transfer of sulfate in plants. SULTRs are associated with growth and development processes and are also involved in stress-dependent processes. Due to the importance of these ion transporters, SULTRs have been studied in some plants, but there is little information available about their function. In the present study, 22 TtSULTR family members were identified and characterized in the genome of Triticumturgidum using available bioinformatics tools. Besides, the exxpression levels of candidate TtSULTR genes were investigated under salt treatments, 150 and 250 mM of NaCl, in different exposed time series. TtSULTRs showed diversity in terms of physiochemical properties, gene structure and pocket sites. TtSULTRs with their orthologues were classified into five main groups and high diversity was observed between SULTR subfamilies. In addition, it was found that segmental duplication events could extended the TtSULTR family members under evolution processes. baxsed on pocket sites analysis, LEU, VAL, and SER amino acids were more detected in the binding site of TtSULTR proteins. Moreover, it was predicted that TtSULTR have high potential to target by phosphorylation modifications. According to promoter site analysis, ABA and MeJA were predicted phytohormones which can more affect the exxpression of TtSULTRs. Real-time PCR analysis revealed that TtSULTRgenes have differential exxpression under 150 mMNaCl, while almost TtSULTRgenes showed similar exxpression patterns in response to 250 mMNaCl and their exxpression levels were reached maximum at 72h after 250 mM salt treatment. Overall, we conclude that TtSULTRgenes are involved in response to salinity in durum wheat. However, more functional studies are needed to determine their precise function and their lixnked-interaction pathways.
