S509 : Effect of growth regulators on in vitro micropropagation of Haworthia spp. plant using leaf explants

Abstarct: Hawothia is a perennial plant in the Liliaceae family and is native to South Africa. Seed propagation of this plant is time consuming and has problems with genetic purity and its non-sexual reproduction through offshoot and cuttings is limited due to the need for abundant native plant, so the use of plant tissue culture can be beneficial for this plant. In this study, the effect of two types of growth regulators of cytokinin BAP and KIN (0.5, 1, 2 mg/l) and two types of growth regulators of auxin IBA and NAA (0, 0.01, 0.1 and 0.5 mg /l) by two type of complete and lower part of leaf explants in modified MS medium were investigated in vitro on Haworthia attenuatae. The results showed that the highest number (3.5), length (1.45 cm) and width (0.87 cm) of shoots were obtained in treatment combination of 0.1 mg/l IBA with 1 mg/l KIN by using the lower part of leaf explant. Complete leaf explants also produced the highest number of shoots (5.22) in combination of 2 mg/l BAP and 0.01 mg/l IBA. Maximum number of leaves (4.66) in 0.5 mg/l BAP treatment without IBA and the longest and widest leaves (1.05 and 0.68 cm) in the highest Levels of cytokinins (BAP and KIN) with 0.5 mg/l IBA were produced using by leaf explants. In this study, the effect of TCL explants thickness (3 and 5 mm) on regeneration of Haworthia plant was also investigated. The highest average of shoot number (1.06) with the highest mean shoot length and width (1 and 0.9 cm) were produced in 0.1 mg/l IBA and 2 mg/l BAP using TCL stem explants with a thickness of 3 mm. In order to produce root, the regenerated shoots were transferred to MS and ½MS media containing IBA and NAA growth regulators (at concentrations of 0, 0.5, 1, 1.5 mg/l). According to the results, the highest number of roots (6.66) were produced in ½MS medium containing 1 mg/l IBA and the highest root length (3.47 cm) was obtained in ½MSmedium with 1.5 mg/l IBA. Finally, rooted plants were transferred to pots containing cocoopeat and perlite at 80% relative humidity and 28-30 °C in order to adaption of greenhouse conditions. Totally, 90% of the Haworthia shoots were adapted to the greenhouse condition.
