S374 : Study on physiological, biochemical and molecular characteristic of tomato lines with low shelf-life

Abstarct: This study was conducted in order to investigate the effect of -MCP and modified atmosphere packaging on some characters of two purified tomato cultivars (Tomato Black cherry (VEG241), Tomato Orange Berry (VEG788) and to evaluate the exxpression profiling of few genes involved in the ethylene biosynthesis. In the current study, tomatoes were harvested at the braker stage from field. Experiment was done as a factorial design in a completely randomized design. The first factor was two tomato lines and the second factor includes film, film with -MCP and control and The third factor consists of four times. The assay was performed in three replications and they were stored at 20oC during 16 days of storage period. The obtained results showed that there were significant differences between control treatment and other mentioned ones in terms of Weight loss, pH, Soluble solids, Firmness and Chlorophyll). In gene exxpression profiling section, gene exxpression pattern of four genes involved in ethylene biosynthesis (ACO1, ACO5, ACS4 and SAM1) were investigated. The gained results demonstrated that the film and film with 1-MCP treatments reduced the exxpression level of mentioned genes compared with that of control. In total, the results showed that the film and film with 1-MCP treatments delayed the he process of the ripening and senescence of tomato fruit.
