S193 : Study of genetic diversity among Azad Shahr Walnuts genotypes (Juglans regia L) using of microsatellite markers.

Abstarct: In order to study genetic diversity of walnut genotypes in Azadshahr, Iran, at first 102 genotypes of four walnut populations (Sayed Abad, Roodbar, Vamian and Kashikar) was assessed with 30 morphological traits (fruit length, fruit diameter, length, etc.). The result of descxription, analysis showed that there are a high diversity among genotypes in some of fruit traits such as kernel percent, fresh and kernel weight of fruits, fruit color, easy kernel separation. The highest average fruit weight (19.79g), observed in Ka17 and Va31 genotypes and Va31 has a highest kernel also (4.9 g). Kernel separation of the fruit in ROOD4 and Va34 genotypes was very easy and they have desirability of fruit flavors index (favorable). The clustering analysis on the baxse of morphological data, genotypes in five distances of cluster divided into eight groups. The fruit and kernel weights have an importance role in of clustering. Geographical populations in this cluster were largely inserted in separate categories. On the baxse of morphological study, 39 genotypes were selected among the genotypes as superior genotypes and genetic diversity of them were investigated by using ten microsatellite markers. In all, this primer was produced 74 polymorphic allele which their sizes ranged between 104 to 267 bp. The number of alleles per locus range from 3 to 11 with the average of 7.4 alleles. The minimum and maximum number of alleles, 3 and 11 alleles, was observed in the WGA005 and WGA054 loci, respectively. The number of effective allele range of 2.026 to 8.974 with the average of 5.80. Dendrogram of population cluster analysis were divided studied populations in two groups. The first group was included Sayed Abad and the second was included Roodbar, Vamian and Kashikar . Cluster analysis of walnut genotypes was made of seven categories. All genotypes collected in the Sayed Abad region were inserted in a cluster and remaining genotypes dispersed in other groups. The result of this study shows that there is a high genetic diversity of walnut in Azadshahr region that can be used by breeders in walnut breeding programs.
