S714 : Effect of methyl jasmonate elicitor on morphological traits, secondary mextabolites and antioxidant enzymes of in vitro Salvia leriifolia

Norozak (Salvia leriifolia Benth.) from the mint family (Lamiaceae) is one of the valuable medicinal plants of the desert regions of Iran, which has medicinal properties such as: antioxidant, antibacterial, antifungal and pain reliever. In this study, the effect of methyl jasmonate (100, 50, 0 and 150 μM) at different times (from seed cultivation to vegetative growth (four weeks, continuous treatment) and one week (pulse treatment) on in vitro seedlings 21 days after cultivation seeds) on in vitro seedlings was investigated by measuring morphological and biochemical traits. The results showed that by increasing the concentration of continuous methyl jasmonate elicitor , morphological traits such as; root length, stem diameter, root beginning diameter, root middle diameter, root end diameter, crown diameter, number of leaves, petiole length, leaf width, fresh and dry weight of root, fresh and dry weight of stem and minimal chlorophyll fluorescence increased and stem length, leaf length and leaf area decreased. By increasing of different stress levels of methyl jasmonate, biochemical traits such as malondialdehyde (MDA), hydrogen peroxide (H2O2), proline, total protein, superoxide dismutase (SOD), catalase, DPPH, iron chelation, nitric oxide, caffeic acid, rosmaric acid, sylvanolic acid, total phenol and flavonoid content increased, and phenylalanine ammonialyase (PAL) and tyrosine ammonialyase (TAL) decreased. By increasing the pulse methyl jasmonate elicitor, morphological traits such as; root length, stem length, stem diameter, root beginning diameter, root middle diameter, root end diameter, crown diameter, petiole length, root fresh and dry weight, and stem fresh weight increased, and stem length, leaf width, leaf length trait, leaf surface, Stem dry weight and minimum chlorophyll fluorescence decreased. Also, biochemical traits such as malondialdehyde (MDA), hydrogen peroxide (H2O2), proline, total protein, superoxide dismutase (SOD), Catalase, DPPH, iron chelation, caffeic acid, rosmaric acid, sylvanolic acid, total phenol and flavonoid content increased, and nitric oxide trapping, phenylalanine ammonialyase (PAL) and tyrosine ammonialyase (TAL) decreased.
