S712 : Effect of Silver nitrate and Silver Nano particles on the morphological traits and antioxidant activities of Salvia Leriifolia Benth under in vitro condition

Abstarct:   Norozak (Salvia leriifolia Benth) is a plant belonging to the mint family (Lamiaceae) Salvia genus, which has very valuable medicinal compounds. In this research, the effect of silver nitrate with concentrations of 0.25, 0.5, and 1 mg/liter, as well as nano silver particles with concentrations of 0.25, 0.5, 1, and 2 mg/liter in different exposure times (continuous and pulsed) were investigated on the in vitro plantlets by measuring the mor phological and biochemical traits. The results showed that using silver nitrate and nanosilver elicitors continuously and pulsed, the morphological traits including leaf length, crown diameter, leaf width, root length, root diameter, root middle diameter, root end diameter, root fresh and dry weight increased compared to the control. It was found that the number of leaves, the fresh weight of the stem and the dry weight of the stem decreased compared to the control. but the leaf area and petiole length did not change significantly compared to the control. Continuous treatments of silver nitrate and nanosilver increased the stem diameter compared to the control Also, the continuous and pulsed treatments of silver nitrate and nano silver increased the biochemical traits including nitric oxide, superoxide dismutase, DPPH, total flavonoid, caffeic acid, rosmarinic acid, salvianolic acid, phenol and proline content compared to the control. Continuous treatments of silver nitrate and nanosilver decreased the activity of phenylalanine ammonia-lyase (PAL) and hydrogen peroxide (H2O2) compared to the control, but increased in a pulsed treatments. The activity of tyrosine ammonia lyase (TAL) showed an increase only in pulsed silver nitrate and nanosilver treatments. The amount of total protein and catalase did not change significantly compared to the control. Malondialdehyde showed an increase only in pulsed silver nitrate and continuous nanosilver treatments, while the amount of iron chelation decreased in all treatments.
