Microorganism with high ability to produce lipid more than 20 percentage of dry weight biomass are called oleaginous microorganism. Mortierell alpine as oleaginous species could grow in media containing glucose and soybean. In this research the appropriated housekeeping genes were determined using RT-qPCR. In initiative of experimentation, soybean and glucose was used to pinpoint the growth rate and lipid production as well as gene exxpression. The results stated that actin gene had the least changes in exxpression in both media. At the rest of experimentation which nano-Mg(OH)2 was used as an additive in previous formulated media was indicated that in soybean medium, constant growth until three days, a steady decline in growth rate from three to four days and after that a fall in biomass accumulation was attained during the rest time. The quantity of lipid during the fermentation was increased and at five days the highest amount of lipid was obtained while in soybean media incorporating nano-Mg(OH)2 biomass production was coincided with lipid production and continued the both growth during the end of fermentation time. Regarding to results of five genes which was analyzed with the bestkeeper and Normv3 software, the presence of nano-Mg(OH)2 caused a steady growth during the five days was observed, in this condition, software analysis indicated that 28 s gene had the best characteristics to select as a best housekeeping gene while at the absence of nano-Mg(OH)2, deterioration of biomass was happened and actin and TEF1were determined as best housekeeping genes.
