S573 : Optimization of indirect organogenesis in Chenopodium quinoa WILD by hypocotyl explant

Abstarct: Quinoa (Chenopodium quinoa Willd.) is considered as a food grain to high nutritional value and as a medicinal plant. Evaluation of quinoa seed germination under light and type of culture medium treatments showed that the highest quinoa seed germination rate(15/87day) was observed in the dark and in MS and ½ MS. Callus formation in quinoa plant using hypocotyl explant on MS medium supplemented with 2,4-D (0, 0.5 and 1 mg/l), nanocarbon (0, 100 and 500 mg/l ) and cytokinin hormones including BAP and KIN (0, 0.25, 0.5 and 1 mg/l) showed that no callus was observed on 2,4-D–free medium. Callus formation by adding BAP showed that the highest percentage of callus formation, weight and area (2.57G and 1.7 Area, respectively) was obtained in medium containing 0.5 mg/l 2,4-D and medium supplemented with BAP produced friable and white callus which was not suitable for regeneration. The cream color and compact calli which selected to organogenesis were produced on the medium supplemented with 1 mg/l 2,4-D, 0.5 mg/l KIN and 500 mg/l nano carbon. Organogenesis in selected calli on MS medium supplemented with both cytokinens BAP and KIN (0, 0.5 and 1 mg/l) in corporated with the three auxins IAA, IBA and NAA (0, 0.1 and 0.2 mg/l), seperately showed that no shoot regeneration was observed in media fee- cytokines. The organogenesis results using IAA showed that the highest shoot regeneration and number of shoot and the longest shoot were observed on medium free-IAA. While the highest length and numbers of roots were obtained at IAA consumption levels. The addition of IBA for organogenesis showed that the highest shoot regeneration and number of shoot and the longest shoot were observed on medium free-IAA, but supplemented with 1 mg/l BAP and KIN. The highest number of roots in this experiment was obtained in medium containing 2 mg/l BAP, 0.2 mg/l IBA and 1 mg/l KIN, while the longest roots were obtained on medium supplemented with 0.5 and 1 mg/l KIN incorporated with 1 mg/l BAP. By adding NAA in organogenesis media showed that the highest shoot regeneration and number of shoot and the longest shoot were obtained on medium supplemented with 1 mg/l BAP and KIN, while the longest and highest number of roots were observed on medium supplemented with 1 mg/l BAP and 0.2 mg/l NAA and 1 mg/l KIN and 0.2 mg/l. In general, the hypocotyl explant can be used to obtain an improved instruction to the indirect micropropagation of quinoa.
