S259 : Effect of Salinity and Drought Stresses on Some Quantitative and Qualitative Characteristics of Nuruozak (Salvia leriifolia Benth) in vitroculture

Abstarct: Nuruozak is valuable plants desert areas of Iran, which has valuable properties such as antioxidant, antibacterial, antifungal and is analgesic. In order to study response of Nurouzak medicinal plant (Salvia leriifolia Benth) to in vitro salinity and drought stresses, two saperate experiments were designed in a completely randomized design with three replications. First test the embryos were cultured on modified MS liquid medium supplemented with different concentrations of at 0, 10, 30 and 50 mM NaCl, as well as PEG at 0, -0.5, -1 and -1.5 bar. The results showed that with increasing levels of salinity and drought, morphological parameters such as leaf area index, chlorophyll, shoot length was significantly decreased (P≤0.01). Also proline, total protein and guaiacol peroxidase activity showed a significant increase compared to the control. In a second experiment to induce callus from growth regular 2,4-D and KIN at concentrations of 0, 5.0: 1, 1: 1, 5.0: 1, 2: 2 was used. The results was showed that medium with 2 mg 2,4-D and 1 mg KIN had the more effective on callus percent of the treatment into account as the most suitable criteria for induction callus was selected. Callus grown on MS medium containing 2 mg 2,4-D and 1 mg KIN after two subcultures, were transferred to salinity and drought environments with similar concentrations in the first experiment. Analysis of datas variance of showed with increasing in salinity and drought catalase activity, guaiacol peroxidase and proline significantly increased ((P≤0.01). While the total protein by increasing salinity and drought significantly decreased. The results of this study suggest that morphological and biochemical characteristics of seedling and callus was affected by both salinity and drought stress.
