S194 : Study on the effects of plant growth regulators on callus induction and plant regeneration in Foeniculum vulgare

Abstarct: Several factorial experiment was done the baxse of completely randomized design for optimizing fennel tissue culture condition. In the first experiment the effect of different concentrations of hormones such as Benzyl amino purine (BAP) and Thidiazuron (TDZ) alone or in combination with different concentrations of naphthalene acetic acid (NAA) and indole butyric acid (IBA) was studied on callus induction hypocotyl and leaf explants in MS medium. The highest callus induction (100%) was observe in 2 mg/l BAP and 0.5 mg/l NAA in hypocotyl explants. In next experiment for shoot regeneration the calli of hypocoltyle and leaf explant cultured in MS medium supplemented with 1 mg/l BAP and 0.2 mg/l NAA. The highest shoot regeneration, 100%, was observe of callus derived from 2 mg/l of BAP and 1 mg/l of NAA. The more the number of regenerated plantlets were transferred io rooting medium containing one milligram per liter of NAA. The highest rooting (75%) was observe of plantlets from callus with combined hormonal 2 milligram per liter BAP,1 milligram per liter NAA.
