S608 : Investigation of the profile of phenolics of Ganoderma lucidum mushroom

Abstarct: Phenolic compounds are one of the most important compounds in most industries, especially the food and drug industry, and are known to have antimicrobial and anti-inflammatory properties. Therefore, today, the extraction of these compounds from different sources, especially plant sources, has attracted much attention of researchers. Also, since infectious diseases cause premature deaths and human breast cancer is one of the deadliest types of cancer in industrialized countries, and considering that the long-term use of commercial antibiotics and currently available chemotherapy agents cause adverse side effects, the increasing demand Society has increased to receive natural anti-bacterial and anti-cancer medicines and substances. This study aims to investigate the phenolic compounds of 3 parts (moss, shell and stem) of Ganoderma lucidum mushroom and to investigate the antibacterial and anticancer effect of the phenolic extract of each 3 parts on 2 species of bacteria Escherichia coli (ATCC25922) and Bacillus subtilis (PTCC1715) and on human breast cancer cells MDA-MB-231 were used. The results showed that the percentage of total phenol in the moss, shell and stem of Ganoderma Lucium mushroom was 2.21, 7.49 and 7.56%, respectively, and the presence of phenolic compounds such as gallic acid, homogenetic acid, chlorogenic acid, p-coumaric acid, O-coumaric acid, protocatechuic acid, malic acid, ferulic acid, p-hydroxybenzoic acid, abscisic acid, catechin, epicatechin, kaempferol, rutin, biochanin A, formonontin, myristicin, naringin, isorhemantin, quercetin, naringenin and hespertin were identified. The minimum inhibitory concentration (MIC) and the minimum bacteriocidal concentration (MBC) of the phenolic extract of all 3 parts of the mushroom were measured by the standard microdilution method, and for Escherichia coli (ATCC25922) the MIC and MBC of the phenolic extract of moss were (250 and 312 µg/ml), shell (375 and 625 μg/ml) and stem (500 and 625 μg/ml) and for Bacillus subtilis (PTCC1715) MIC and MBC of phenolic extract of moss (2500 and 3000 μg/ml), shell (3000 and 4000 μg/ml) ml) and stem (3000 and 4000 µg/ml). Also, the effect of the cytotoxicity of the phenolic extract of 3 parts of the mushroom on the human breast cancer cell line MDA-MB-231 was measured by the MTT cytotoxicity test, and the results showed that the phenolic extract of the stem and shell of the mushroom significantly increased cell death at a dose of 500 μg/ml and the IC50 levels of shell and stem were 465.3 and 485.7 μg/ml, while moss did not reach 50% inhibition.
