S198 : The effects of auxins and Agrobacterium rizhogenes on rooting of YeW cuttings (Taxus baccata)

Abstarct: A greenhous experiment was conducted to evaluate the effects of strains agrobacterium rhizogenes and some ouxins(IBA andNAA) on adventitious root formation in stem cutting of Taxus bacata L. fifteen to twenty centimeter long stem cutting treated with different hormones and strains agrobacterium rhizogenes. Were transplanted in substrate containing ½ peat and ½ perlite and were kept in a greenhouse at 18-25 c. day/night temperatures. The treatments were using strains A4, LB9402, C58C2 with OD=0.6 and hormones IBA, NAA in differents concentrations 0, 500, 2500 and 0, 500, 2500 , 5000 ppm respectively and comparing them to control. The results indicated that positive effect of A4, LB9402 and IBA , NAA treatments on percentation Root formation, callus formation, root number,root medium length, increase root length and lateral. root number. Strains A4 , LB9402 in present IBA and NAA alone and IBA , NAA in combination together were significantly effect on Root formation, callus formation, root number,root medium length, increase root length and lateral root number. The results showed that using strains A4, LB9402 in present both hormones IBA , NAA in different concentrations 5000 and 2500 ppm respectively resulted the highest percentation root formation(90/66% , 87/5% respectively) and callus formation.
