GV277 : The effect of eight weeks of endurance training with garlic and stevia supplementation on the exxpression of genes involved in energy mextabolism located in the hypothalamic arch "POMC" and "MC4R" of obese male Wistar rats

Abstarct: Background and Aim: Studies on the effect of exercise and dietary supplements on the exxpression of hypothalamic genes involved in appetite and low obesity. Therefore, the aim of this study was to investigate the effect of eight weeks of endurance training with garlic and stevia supplementation on the exxpression of genes involved in hypothalamic arch mextabolism (POMC and MC4R) in obese male Wistar rats. Methods: In the present experimental study, 35 adult Wistar rats were divided into seven groups. Six of them underwent a high-calorie diet to gain weight, and one group followed a normal diet. The groups were: exercise-obesity, garlic-obesity supplement, stevia-obesity supplement, garlic-obesity garlic supplement and exercise, stevia-obesity and obesity supplement and control. The rats received stevia supplementation of 250 mg per kg body weight of rats and the garlic supplement group received 100 mg / kg body weight of rats for 5 days a week for eight weeks. Endurance training program was performed on the treadmill for 30 minutes a day, 8 meters per minute and 5 days a week. At the end, the head was removed from the trunk by a guillotine and then immediately removed by splitting the skull of the brain. The ventral surface of the brain was placed upwards and a 4 mm thick incision containing the hypothalamus was prepared POMC and MC4R. Data were analyzed using ANOVA and LSD post hoc test by SPSS software version 19 at a significance level of 0.05. Results: The results showed that only between the control group and the obese and exercise groups with garlic there was a significant difference in POMC gene exxpression (P <0.05). MC4R gene exxpression also increased in all obese groups that received the intervention, but this increase was not statistically significant (P˃0.05). Conclusion: baxsed on the findings of this study, endurance training, garlic and stevia supplementation increased the exxpression of genes involved in hypothalamic arch mextabolism (POMC and MC4R) and subsequently decreased appetite, but due to inconsistent results with some studies The former needs further study in this area.
