S200 : Effect of phytohormones tissue and regeneration of lime (citrus aurantifolia)

Abstarct: This research was carried out in three parts; Indirect regeneration node and internodes of Lisbon lime and two ecotypes of Mexican lime (namely, hormozgan and jahroum) in MS and MT media culture containing 2 mg/l 2,4-D. Indirect regeneration of epicotyl, leaf and cotyledonary leaf of the same genotype and ecotypes in MS media culture with thirteen hormonal combinations, and direct regeneration of epicotyl, leaf and cotyledonary leaf in MS with twelve hormonal combinations. Unfortunately, the first experiment was failed because of bacterial infection of produced calli. In the second experiment, explant treated with 1, 2 mg/l 2,4-D, 1, 2, 3 mg/l 2,4-D + 1mg/l kinetin produced calli. The variance was demonstrated that genotype, explant and hormone had significant effect on callus formation. Furthermore, friable and no friable calli were formed. The produced calli were moved in to photoperiod condition: light to induce shoot formation and dark to induce embryo generation. After 8 weeks of incubation, no changes in calli was observed, except for few that turned green. In the third experiment, hormone and explant had significant effect on shoot formation.
